Design Notes

For hsa-miR-30e-5p, we designed multiple synthetic sequences called Toehold Switches, which have the ability to bind with a part of the miRNA molecule. Furthermore, in order for the required thermodynamic conditions to be met, the hairpin-like structure of the toeholds was designed to lack the pre-RBS area in the loop and to maintain strong bonds between the paired bases of the body of the hairpin.

This part was designed by the members of our Dry Lab team using NUPACK and a function they coded in Python [ standard_design() ] and it was synthesized by IDT (Integrated DNA Technology).
